Suppose I book a tatkal ticket in Siliguri from PRS...
Can I get the same ticket in kolkata by telling the PRS counter officer that I lost my ticket...?????

No duplicate Tatkal tickets shall be issued. Duplicate Tatkal tickets shall be issued only in exceptional cases on payment of full fare including Tatkal charges.
There is no ememptional cases...
There is no dupilicate tatkal tickets.
My friend lost his Tatkal ticket. He was not even able to get a duplicate inspite his mom being the PRS RS.

The Words mentioned are written in rules dude i am not posting it by myself :)
Duplicate Tatkal Tickets are usually not issued unless and until the situation is extreme and you can convince the higher officials regarding the same. Consider it as 99% No.
